Bioconversion of hydrolyzed cashew peduncle bagasse for ethanol and xylitol production AGRIAMBI
Medeiros,Lorena L. de; Silva,Flávio L. H. da; Santos,Sharline F. M.; Madruga,Marta S.; Melo,Débora J. N. de; Conrado,Líbia de S..
ABSTRACT The agro-industrial waste deposited in the environment causes problems in nature that can be solved with the use and generation of bioproducts. Thus, the objective was to study the lignocellulosic fraction of cashew (Anacardium occidentale L.) peduncle bagasse and fermentation on large scale (8-16 times) using the strain Candida guilliermondii CCT-3544 as production agent. According to the obtained results, it can be noted that the dry cashew peduncle bagasse has 21.45% of cellulose, 10.96% of hemicellulose and 35.39% of lignin. During fermentation, C. guilliermondii 3544-CAT was able to grow on medium containing hydrolysate, with maximum cell growth concentration of 3.5 g L-1. The behavior of the sugars in the fermentation process was similar in...

Cell wall biochemistry of sapodilla (Manilkara zapota) submitted to 1-methylcyclopropene Braz. J. Plant Physiol.
Morais,Patrícia L.D.; Miranda,Maria R.A.; Lima,Luis C.O.; Alves,José D.; Alves,Ricardo E.; Silva,José D..
Sapodilla (Manilkara zapota) is a climacteric fruit that ripens shortly after harvest. Studies on its conservation during storage have been mainly restricted to using low temperatures and modified atmospheres. In this study we investigated the influence of 1-methylcyclopropene (1-MCP) on the physiological and biochemical changes that sapodilla cell wall undergoes during ripening and evaluated its potential to preserve sapodilla fruits at postharvest. Fruits were treated with ethylene antagonist 1-MCP at 300 nL L-1 for 12 h and then stored under a modified atmosphere at 25ºC for 23 d. 1-MCP significantly delayed softening of sapodilla for 11 d as a consequence of inhibition of cell wall degrading enzyme activities, and thus 1-MCP-treated fruit exhibited a...

Cloning and endogenous expression of a Eucalyptus grandis UDP-glucose dehydrogenase cDNA Genet. Mol. Biol.
Labate,Mônica T. Veneziano; Bertolo,Ana L. Ferreira; Nascimento,Daniela Defávari do; Gutmanis,Gunta; Andrade,Alexander de; Rodrigues,Maria J. Calderan; Camargo,Eduardo L.O.; Boaretto,Luis Felipe; Moon,David H.; Bragatto,Juliano; Labate,Carlos A..
UDP-glucose dehydrogenase (UGDH) catalyzes the oxidation of UDP-glucose (UDP-Glc) to UDP-glucuronate (UDP-GlcA), a key sugar nucleotide involved in the biosynthesis of plant cell wall polysaccharides. A full-length cDNA fragment coding for UGDH was cloned from the cambial region of 6-month-old E. grandis saplings by RT-PCR. The 1443-bp-ORF encodes a protein of 480 amino acids with a predicted molecular weight of 53 kDa. The recombinant protein expressed in Escherichia coli catalyzed the conversion of UDP-Glc to UDP-GlcA, confirming that the cloned cDNA encodes UGDH. The deduced amino acid sequence of the cDNA showed a high degree of identity with UGDH from several plant species. The Southern blot assay indicated that more than one copy of UGDH is present...

Draft genome of Thermomonospora sp. CIT 1 (Thermomonosporaceae) and in silico evidence of its functional role in filter cake biomass deconstruction Genet. Mol. Biol.
Omori,Wellington P.; Pinheiro,Daniel G.; Kishi,Luciano T.; Fernandes,Camila C.; Fernandes,Gabriela C.; Gomes-Pepe,Elisângela S.; Pavani,Claudio D.; Lemos,Eliana G. de M.; Souza,Jackson A. M. de.
Abstract The filter cake from sugar cane processing is rich in organic matter and nutrients, which favors the proliferation of microorganisms with potential to deconstruct plant biomass. From the metagenomic data of this material, we assembled a draft genome that was phylogenetically related to Thermomonospora curvata DSM 43183, which shows the functional and ecological importance of this bacterium in the filter cake. Thermomonospora is a gram-positive bacterium that produces cellulases in compost, and it can survive temperatures of 60 ºC. We identified a complete set of biomass depolymerizing enzymes in the draft genome of Thermomonospora sp. CIT 1, such as α-amylase, catalase-peroxidases, β-mannanase, and arabinanase, demonstrating the potential of this...

Flooding tolerance and cell wall alterations in maize mesocotyl during hypoxia PAB
Vitorino,Patrícia Goulart; Alves,José Donizeti; Magalhães,Paulo César; Magalhães,Marcelo Murad; Lima,Luíz Carlos Oliveira; Oliveira,Luiz Edson Mota de.
This research aimed to characterize the tolerance to flooding and alterations in pectic and hemicellulose fractions from mesocotyl of maize tolerant to flooding when submitted to hypoxia. In order to characterize tolerance seeds from maize cultivars Saracura BRS-4154 and BR 107 tolerant and sensitive to low oxygen levels, respectively, were set to germinate. Plantlet survival was evaluated during five days after having been submitted to hypoxia. After fractionation with ammonium oxalate 0.5% (w/v) and KOH 2M and 4M, Saracura BRS-4154 cell wall was obtained from mesocotyl segments with different damage intensities caused by oxygen deficiency exposure. The cell wall fractions were analyzed by gel filtration and gas chromatography, and also by Infrared...

Kinetics of high-Level of ß-glucosidase production by a 2-deoxyglucose-resistant mutant of Humicola lanuginosa in submerged fermentation BJM
Bokhari,Syed Ali Imran; Latif,Farooq; Rajoka,Muhammad Ibrahim.
A 2-deoxyglucose-resistant mutant (M7) of Humicola lanuginosa was obtained by exposing conidia to γ-rays and permitting expression in broth containing 0.6% 2-deoxyglucose (DG) and cellobiose (1%) before plating on DG esculin-ferric ammonium citrate agar medium from which colonies showing faster and bigger blackening zones were selected. Kinetic parameters for enhanced ß-glucosidase (BGL) synthesis by M7 were achieved when corncobs acted as the carbon source. The combination between corncobs and corn steep liquor was the best to support higher values of all product formation kinetic parameters. Effect of temperature on the kinetic and thermodynamic attributes of BGL production equilibrium in the wild organismand M7was studied using batch process at eight...

Xylose production from sugarcane bagasse by surface response methodology AGRIAMBI
Paiva,José E. de; Maldonade,Iriani R.; Scamparini,Adilma R. P..
The aim of this study was to optimize the production of xylose from sugarcane bagasse. The hydrolysis process was carried out to evaluate the effect of temperature and sulphuric acid concentration on the xylose production at 18, 30, and 50 min of hydrolysis. A Central Composite Rotatable Design (CCRD) was used to study two process variables, temperature (111.5; 114.0; 120.0; 126.0 and 128.5 ºC) and sulphuric acid concentration (0.20; 0.70; 1.90; 3.10 and 3.60% v v-1). Sulphuric acid had a greater influence on the hydrolysis process than temperature. At concentrations of sulphuric acid higher than 3.10% (v v-1), raising temperature had little influence on the yield of xylose production. The best condition to obtain xylose (266.73 mg g-1 of dry bagasse) was...
